Toss-up between The Breakfast Club and Ferris Bueller's Day Off.
The Breakfast Club had characters that I identified with more, but one aspect of the ending -- Ally Sheedy's makover -- just fell flat for me.
Whereas Ferris Bueller had characters that I wanted to be but couldn't actually identify with, but had an aspect of the ending (Cameron's determination to stand up to his father in the face of very real fear) that just absolutely sang to me.
I just realized that I've never actually seen Planes, Trains, and Automobiles. I'll need to correct that.
"Some Kind of Wonderful." It had the ending I wanted from "Pretty in Pink," terrific performances from Mary Stuart Masterson and Eric Stoltz, and Duncan. 'Nuf said.

Ferris was great but I loved SIXTEEN CANDLES. Every moment was histerical. But the subtle things make me giggle the most. Joan Cusack in a back brace trying to get a drink at a drinking fountain, etc. All were fabulous! Its a real sad day!

My fave JH would have been FBDO, but I also learned in the comments that he did Planes, Trains, and Automobiles which was very good. That is my favorite.
I've never seen PiP or Sixteen Candles so I'm only basing this on the ones I *have* seen so: Some Kind of Wonderful. I fell in love with both women and have followed their careers for 20+ years....
Some Kind of Wonderful. I re-wound and rewatched that kiss (you know the one) so many times I wore out the tape on the VHS. Every time I hear "She Loves Me" my mind goes right back to that scene.
Having trouble choosing just one... but while thinking it over I think Pretty In Pink has shaped my life in ways I don't acknowledge--my obsessive love of Duckie, love of Annie Potts' character... but it also disappoints in so many ways (the complete ending, Annie Potts' character turning square and going out with the pet shop guy...)

Parker Lewis came close but that was played too much like a romp to be believable. Ferris could have done all that weird stuff, y'know?
Uncle Buck is my favourite. If you watch it carefully it's all the components of Uncle Buck being a homicidal maniac or at the least some type of serial killer. How else do you explain giant pancakes with a fresh shovel? How else do you explain invading a rowdy teenage party, drilling the lock, and smuggling a teenager out without anyone noticing he's being put into the trunk of a car? Everyone knows he's a maniac they are just too scared to say anything because he hasn't gotten caught. Oh that John Hugh's was so sly to slip that in without anyone noticing.
My 21 year old niece recently discovered The Breakfast Club, which reminded me of how much I'd loved Some Kind of Wonderful back in the day. I had such fond memories that I purchased it on DVD just so I could have it.

I agree with the guy who said Career Opportunities is underrated. I love that movie, I think if the store robbery plot were removed you'd have an amazing indie dramady. Weirdly out of all Hughes films I've probably seem it the most often. Since I first saw it in theatres in 91 I've probably watched it 50 times.

But looking at John Hughes's imbd page everything after Uncle Buck is a wasteland.

Ferris just barely inches out The Breakfast Club, but mainly for the genius in disguising a movie about Cameron as a movie about Ferris.
I also have a great deal of love for that scene in Uncle Buck where he scares the pants of that boy with the drill.
